Response surface methodology (RSM) was applied to optimise the experimental conditions for ultrasound-assisted extraction (UAE) to maximise the recovery of total extraction yield (TEY), total polyphenol content (TPPC), total flavonoid content (TFC), and total tannin content (TTC) from Crataegus fruit (CF). A three-factor (ethanol concentration (20 - 80%), sonication time (10 - 40 min) and temperature (20 - 60 °C) and three-level (-1, 0, and +1) Box-Behnken design (BBD) was applied. Ethanol concentration and sonication temperature were the major factors influencing the recovery of TEY , TPPC, TFC and TTC from CF . At the optimal UAE conditions (ethanol concentration 54%, duration 27 min and temperature 43 °C), the experimental value of TPPC was 69.83 mg gallic acid equivalents (mg GAE/g CFE), well matched with the predicted value of 70.5 mg GAE/g, which confirming the successful application of the response surface methodology (RSM) model. UAE showed higher extraction yield, (TPPC, TFC, TTC), antioxidant activity (DPPH, FRAP , β-carotene, phosphomolybdate),and anti-inflammatory activity (chicken albumin assay) than conventional ethanol extraction. This study identified the ideal UAE settings for the maximum recovery of bioactive compounds from CF that could be used as natural antioxidant agent or functional ingredient enhancers and as therapeutic bioactive extract. |
